She's a sannin because of her amazing medical ninjutsu and strength. Not for her fantastic fighting skills.

I reckon Rock Lee would beat her without his weights on. :p

With his speed, Gai would easily beat her, until she calls out her slug. Then two or three gates would finish it.

I think Gai would simply be too fast for Tsunade to catch and so he would win with sheer speed.
